Escalation of anthropogenic load on the biosphere is accompanied by increasing use of pesticides of various chemical groups, among which phenoxyacetic acid derivatives, in particular 2,4-dichlorophenoxyacetic acid, are among the most widely used. The aim of the study was to assess morphological changes in the liver and testes of rats under the influence of chronic exposure to 2,4-dichlorophenoxyacetic acid (2,4-DA) and the effectiveness of correction with mexidol. Three experimental groups were formed: intact group (healthy animals, no interventions) – 10 animals; 2,4-DA group (30 days) – toxic injury model; 2,4-DA + mexidol group (30 days), 10 rats. During the experiment, daily clinical monitoring of the animals was performed (body weight, behavior, coat condition, appetite). To analyze structural changes in the organs, the method of semithin sections and hematoxylin-eosin staining were used, with examination by light microscopy. Chronic administration of 2,4-DA caused pronounced dystrophic and inflammatory changes in the liver, including disorganization of the plate structure, hepatocyte vacuolization, nuclear pyknosis, sinusoidal congestion, infiltration of the portal tracts and disruption of the structure of the bile ducts. In the testes, disorganization of the epitheliospermatogenic layer, lower representation of spermatogonia, vacuolization of spermatids and microcirculatory disorders were observed, accompanied by lymphoid infiltration and decreased mitotic activity, which indicated suppression of spermatogenesis and a toxic effect on reproductive potential. The use of mexidol against the background of 2,4-DA administration was accompanied by restoration of structural elements of the examined organs: in the liver, alignment of hepatocyte shape, decreased infiltration, restoration of circulatory relationships in the sinusoids and signs of activation of reticuloendothelial cells were observed, reflecting activation of protective and detoxification mechanisms; in testicular tissues, the frequency of gross defects of epithelial stratification and basement membrane detachment decreased, the number of vacuolized spermatids was reduced, but mitotic figures among spermatogonia occurred rarely. The obtained results indicate that 2,4-DA causes systemic morphological disorders in the liver and testes associated with oxidative stress and inflammatory-dystrophic reactions, while mexidol shows a positive tendency toward recovery, more pronounced in liver tissue and less pronounced in the testes.
